The league leaders announced that the Dutch player had a "fever" in a statement, while coach Hansi Flick revealed on Monday that the Uruguayan defender Ronald Araujo would be absent from the match due to a "personal matter".
Araujo was shown a red card in Barcelona's 3-0 Champions League loss to Chelsea last week, and Spanish media have reported that he has requested a "mental health break".
"Ronald is currently not ready," Flick told the press on Monday."It’s a private matter. I don’t want to elaborate. Please respect that."
Both De Jong and Araujo missed Saturday’s league victory over Alaves, the former due to a family issue, and the latter because of a stomach illness, Barcelona stated.
The Catalan side is also without Fermin Lopez alongside long-term absentees Marc-Andre ter Stegen and Gavi.
Current champions Barca lead by three points over Atletico, who are in fourth place, ahead of their meeting at Camp Nou.
